<p>Établie à 430 mètres d'altitude<sup id="cite_ref-1" class="reference"><a href="#cite_note-1"><span class="cite-bracket">[</span>1<span class="cite-bracket">]</span></a></sup>, la gare d'Amplepuis est située au <a href="Point_kilom%C3%A9trique" title="Point kilométrique">point kilométrique</a> (PK) 448,780 de la <a href="Ligne_du_Coteau_%C3%A0_Saint-Germain-au-Mont-d'Or" title="Ligne du Coteau à Saint-Germain-au-Mont-d'Or">ligne du Coteau à Saint-Germain-au-Mont-d'Or</a> entre les gares de <a href="Gare_de_Saint-Victor_-_Thizy" title="Gare de Saint-Victor - Thizy">Saint-Victor - Thizy</a> et <a href="Gare_de_Tarare" title="Gare de Tarare">Tarare</a>. La gare dispose de deux quais latéraux qui mesurent 213 <abbr class="abbr" title="mètre">m</abbr><sup id="cite_ref-2" class="reference"><a href="#cite_note-2"><span class="cite-bracket">[</span>2<span class="cite-bracket">]</span></a></sup>.

<p>La gare d'Amplepuis est mise en service en <time class="nowrap date-lien" datetime="1866" data-sort-value="1866"><a href="1866_dans_les_chemins_de_fer" title="1866 dans les chemins de fer">1866</a></time> par la <a href="Compagnie_des_chemins_de_fer_de_Paris_%C3%A0_Lyon_et_%C3%A0_la_M%C3%A9diterran%C3%A9e" title="Compagnie des chemins de fer de Paris à Lyon et à la Méditerranée">Compagnie des chemins de fer de Paris à Lyon et à la Méditerranée</a> (PLM). Elle est une gare en cul-de-sac jusqu'à la mise en service du <a href="Tunnel_des_Sauvages" title="Tunnel des Sauvages">tunnel des Sauvages</a> de la section comprise entre Amplepuis et Tarare le <time class="nowrap" datetime="1868-10-19" data-sort-value="1868-10-19">19 octobre 1868</time><sup id="cite_ref-3" class="reference"><a href="#cite_note-3"><span class="cite-bracket">[</span>3<span class="cite-bracket">]</span></a></sup> complétant la ligne dans son intégralité.

Elle est équipée d'automates pour l'achat de titres de transport TER<sup id="cite_ref-SNCFTERGare_5-0" class="reference"><a href="#cite_note-SNCFTERGare-5"><span class="cite-bracket">[</span>5<span class="cite-bracket">]</span></a></sup>.
